I don't really follow hamstuck, so I don't know who the sufferer is, and I don't understand what "Check you privilege" means.
I assume it's either a reference or a joke of some sort.

No you pretty much do need to read Homestuck to know who any of the characters are
It's a popular webcomic but it's not that popular

"Check your privilege" is not really said by anyone who isn't on Tumblr either, there's no so much "social justice" presence on most media platforms.
